PDE5 inhibitors are usually sorted by duration alone. Tadalafil’s unrelated scaffold produces an inverted selectivity profile: it spares the retinal enzyme PDE6 while potently inhibiting PDE11 – and the reported PDE11 potency turns out to depend on which splice variant was assayed.
